Sestrin prevents atrophy of disused and aging muscles by integrating anabolic and catabolic signals

Ageing is associated with muscle atrophy, which negatively impacts quality of life. Here the authors show that expression of sestrins decreases during inactivity and that their overexpression prevents atrophy in mice via modulation of autophagy and protein degradation.

Geriatric muscle stem cells switch reversible quiescence into senescence

This study shows that ageing satellite cells undergo an irreversible transition from a quiescent to a pre-senescent state that results in the loss of muscle regeneration in sarcopenia; furthermore, increased expression of p16^INK4a is identified as a common feature of senescent satellite cells.

Skeletal muscle stem cells (or satellite cells, SCs) are normally quiescent but activate and expand in response to injury. Here the authors show that induction of DHX36 helicase during SC activation promotes mRNA translation by binding to 5′UTR mRNA G-quadruplexes (rG4) in targets including Gnai2 and unwinding them.

The regenerative properties of muscle stem cells decline with age as the stem cells enter an irreversible state of senescence; a study of mouse muscle stem cells reveals that entry into senescence is an autophagy-dependent process and promoting autophagy in old satellite cells can reverse senescence and restore their regenerative properties in an injury model.

3-Deazaadenosine alleviates senescence to promote cellular fitness and cell therapy efficiency in mice

With age, muscle stem cells have been reported to undergo a quiescence-to-senescence switch, thus reducing regenerative capacity. Here, using in vitro and mouse models, Guerrero et al. report that treatment with 3-deazaadenosine alleviates senescence and preserves the regenerative potential of muscle and hematopoietic stem cells.
