ERRα mediates metabolic adaptations driving lapatinib resistance in breast cancer

Despite initial benefits in treating HER2-positive breast cancer patients with lapatinib, resistance is prevalent. Here the authors show that lapatinib resistance can be ascribed to mTOR-mediated re-activation of ERRα and to the consequent induction of a metabolic adaptation.

GLUT1 inhibition blocks growth of RB1-positive triple negative breast cancer

Triple negative breast cancer is a deadly form of breast cancer with limited therapeutic options. Here the authors show the efficacy of GLUT1 pharmacological inhibition against a subset of tumors expressing RB1, thereby identifying RB1 protein level as a biomarker of sensitivity to anti-GLUT1 therapy.

Convalescent plasma for hospitalized patients with COVID-19: an open-label, randomized controlled trial

A randomized trial in patients hospitalized with COVID-19 showed no benefit and potentially increased harm associated with the use of convalescent plasma, with subgroup analyses suggesting that the antibody profile in donor plasma is critical in determining clinical outcomes.

Oestrogen-related receptors (ERRs) have been shown to control vast gene networks that are involved in glycolysis, glutaminolysis, oxidative phosphorylation, nutrient sensing and biosynthesis pathways. This Review discusses these findings in the context of breast cancer and discusses whether targeting the ERRs for the development of cancer therapeutics is feasible.
