E3 ubiquitin ligase RNF10 promotes dissociation of stalled ribosomes and responds to ribosomal subunit imbalance

Perturbation of protein synthesis causes ribosome stalling on mRNAs. Here, Lehmann et al. demonstrate that the E3 ubiquitin ligase RNF10, through monoubiquitination of RPS3, promotes dissociation of the small ribosomal subunit upon ribosome stalling.

RNF219 attenuates global mRNA decay through inhibition of CCR4-NOT complex-mediated deadenylation

The CCR4-NOT complex is a key regulator of eukaryotic mRNA deadenylation. Here the authors identified the E3 ubiquitin ligase RNF219 as an acetylation-regulated cofactor of the complex, which inhibits mRNA decay through a direct interaction with NOT9.

RNA cytosine methylation by Dnmt2 and NSun2 promotes tRNA stability and protein synthesis

Although cytosine-C5 methylation is a prominent modification of tRNAs, its functional significance has been unclear. Mice that lack both the Dnmt2 and NSun2 tRNA methyltransferases showed developmental and cellular differentiation defects, and loss of Dnmt2 and NSun2 was further associated with tRNA degradation and reduced rates of protein synthesis, suggesting that this modification promotes mouse development by supporting protein synthesis.

Christoph Plass and colleagues investigate the transcriptomic and epigenomic changes induced by treatment with inhibitors of DNMT and HDAC in cancer cell lines. They observe large numbers of treatment-induced non-annotated TSSs (TINATs) encoded in long-terminal repeats that are normally repressed in most cell types.

Structures in 5′ untranslated regions of eukaryotic mRNAs contribute to gene regulation by controlling cap-dependent and cap-independent translation initiation through diverse mechanisms. New structure probing technologies coupled with techniques such as compensatory mutagenesis will likely identify new structured RNA elements and help elucidate their function.
