Engineered IscB–ωRNA system with expanded target range for base editing

Xiao et al. characterize 19 previously unidentified IscB proteins, 10 of which show activity in mammalian cells. They engineer one of the proteins into a robust in vivo genome-editing tool with a broad target-adjacent motif scope and further develop it into a base editor.

Alternative lengthening of telomeres (ALT) provides cancer cells a mechanism to sustain replicative immortality. Here, the authors identify KDM2A as a molecular vulnerability in ALT-dependent cancer cells and demonstrate its role in the resolution of ALT-specific telomere clusters via recruitment of SENP6.

Glucocorticoid receptor regulates PD-L1 and MHC-I in pancreatic cancer cells to promote immune evasion and immunotherapy resistance

Glucocorticoids and glucocorticoid receptor (GR) signalling can suppress anti-tumour immunity. Here the authors show that GR activates PD-L1 expression and represses MHC-I expression in pancreatic cancer cells, while GR inhibition enhances anti-tumour immunity and sensitises the cancer cells to immunotherapy.

Kras activation and p16 inactivation cooperatively promote pancreatic cancer progression.
