Therapeutic antibodies represent a powerful tool for immunotherapy but to design their properties to make them functionally versatile but similar to naturally produced human antibodies is challenging. Here the authors engineer human B cells to express heavy chain only antibodies with mutations to avoid pairing with endogenous antibodies and to enhance effector function and durability.
