Morphogenetic gradients can emerge through the slowdown of morphogen diffusion, however, the precise monitoring of this process across micrometre scales remains difficult. Here, the authors present Single-Plane Illumination Microscopy-based spatial Fluorescence Cross-Correlation Spectroscopy (SPIM-sFCCS) to measure the diffusion of the morphogen Squint in early zebrafish embryos as a function of topography and length scale.
