Labelizer: systematic selection of protein residues for covalent fluorophore labeling

Gebhardt and colleagues developed a computational method using a naïve Bayes classifier to identify optimal protein labelling sites. Their analysis of 100+ proteins revealed four predictive parameters, leading to a Python package and a web-tool for protein structure analysis and labelling score calculations.

First insights into the biodiversity and biogeography of the Southern Ocean deep sea

Three sampling expeditions of the Southern Ocean provide the first description of its tremendous biodiversity, including 585 isopod species new to science, and clarifies patterns of species dispersion. Previously this ocean was not thought to be particularly rich.

Structure of the haptoglobin–haemoglobin complex

This study reports the crystal structure of porcine haptoglobin in complex with haemoglobin at 2.9 Å resolution; this provides a structural basis of haptoglobin-mediated recognition of haemoglobin, and insight into the protective role of haptoglobin at the atomic level.

Global kinome profiling reveals DYRK1A as critical activator of the human mitochondrial import machinery

Mitochondrial protein import is mediated by the translocase of the outer membrane (TOM), through which nearly all precursors traverse. Here, the authors perform global in vitro kinome profiling and by this identify that DYRK1A phosphorylates TOM70 and promotes import.

Reverse optogenetics of G protein signaling by zebrafish non-visual opsin Opn7b for synchronization of neuronal networks

Microbial rhodopsins can be used to control action potentials, while animal opsins can be used to control intracellular signaling pathways. The authors identify Opn7b as constitutively active G_i/o coupled receptor that can be deactivated by light and used to modulate neuronal activity.

In the non-randomized phase 3b/4 DESTINY-Breast12 study, trastuzumab deruxtecan (T-DXd) treatment of patients with HER2⁺ advanced breast cancer and active or stable brain metastases showed consistent intracranial activity and systemic efficacy of T-DXd.
