Impaired thermogenesis and adipose tissue development in mice with fat-specific disruption of insulin and IGF-1 signalling

Insulin and insulin-like growth factor 1 signalling pathways have overlapping effects on adipose tissue and glucose homeostasis. Boucheret al.created fat-specific double knockouts of these pathways and demonstrated their crucial role for adipocyte development, metabolism and thermogenesis in mice.

Ablation of TRIP-Br2, a regulator of fat lipolysis, thermogenesis and oxidative metabolism, prevents diet-induced obesity and insulin resistance

TRIP-Br2–null mice are resistant to obesity and insulin resistance and have higher energy expenditure because of increased thermogenesis and oxidative metabolism. As expression of this transcriptional regulator is elevated in fat from obese humans, TRIP-Br2 might be a new therapeutic target against insulin resistance and hyperlipidemia.

A regulatory subunit of phosphoinositide 3-kinase increases the nuclear accumulation of X-box–binding protein-1 to modulate the unfolded protein response

Insulin signals through phosphoinositide 3-kinase (PI3K) to induce cell growth, which often increases protein translation and stress of the endoplasmic reticulum (ER). In two new studies, Umut Ozcan and Ron Kahn and their colleagues now find that in response to insulin signaling, p85α and p85β, the heterodimeric regulatory subunits of PI3K, can increase the nuclear localization of a key transcription factor that resolves ER stress (pages 374–376 and pages 429–437).

Domain-dependent effects of insulin and IGF-1 receptors on signalling and gene expression

Despite being structurally similar, the insulin receptor (IR) and insulin growth factor I receptor (IGF1R) elicit distinct signalling pathways. Liver macrophages regulate systemic metabolism through non-inflammatory factors

Proinflammatory activation of liver macrophages and their secretion of proinflammatory cytokines have been linked to obesity. Here Morgantini et al. report a mechanism through which liver macrophages can impair liver metabolism and promote insulin resistance in obesity in the absence of an overt proinflammatory phenotype, through secretion of non-inflammatory factors such as IGFBP7.
