Fasting boosts breast cancer therapy efficacy via glucocorticoid activation

Beneficial effects of fasting combined with endocrine therapy for oestrogen receptor-α-expressing breast cancers can be recapitulated using exogenous glucocorticoid receptor ligands instead of fasting to reduce harmful effects.

Tailored cobalt-salen complexes enable electrocatalytic intramolecular allylic C–H functionalizations

Oxidative allylic C–H functionalizations minimise the need for functional group activation and generate alkenyl-substituted products amenable to further chemical modifications. Here the authors report an oxidant-free, electrocatalytic approach to achieve intramolecular oxidative allylic C–H amination and alkylation by employing tailored cobalt-salen complexes as catalysts.

Deleted in colorectal carcinoma suppresses metastasis in p53-deficient mammary tumours

In a mouse model of mammary carcinoma, loss of deleted in colorectal cancer (DCC) promotes metastasis formation, and in cell cultures derived from p53-deficient mouse mammary tumours DCC expression controls netrin-1-dependent cell survival, supporting the function of DCC as a context-dependent tumour suppressor that limits survival of disseminated tumour cells.

p15^Ink4b is a critical tumour suppressor in the absence of p16^Ink4a

A locus on chromosome 9p21 which encodes three cell cycle inhibitors is frequently deleted in human cancer. Two of these, p16^INKa and p14^ARF, function as tumour suppressors. A new mouse model now shows that a third, p15^INK4b, also contributes to the tumour suppressor function of this locus. Mice lacking all three show enhanced tumourigenesis and a broader tumour spectrum.

A mouse model that is immunologically tolerant to reporter and modifier proteins

Bresser and Dijkgraaf et al. develop the ‘Tol’ strain, a genetically modified mouse model that expresses a range of shuffled reporter and modifier proteins from a single open reading frame. This strain is immunologically tolerant to these reporter and modifier proteins, providing a valuable model system for cell transfer studies and virus-mediated gene transfer studies.

Lethal graft-versus-host disease in mouse models of T cell receptor gene therapy

T cell receptor (TCR) gene therapy is a promising immunotherapy for cancer and infectious disease. But introducing exogenous TCR α and β chains into T cells may have unintended consequences. In this issue, Bendle et al. show that the transfer of TCR-transduced T cells into mice triggered a lethal pathology that resembles graft-versus-host disease and is caused by the pairing of endogenous and exogenous TCR chains resulting in autoreactive T cells (pages 520–521).

Cooperative targeting of melanoma heterogeneity with an AXL antibody-drug conjugate and BRAF/MEK inhibitors

Expression of AXL earmarks melanoma cells resistant to BRAF and MEK inhibitors that either pre-exist in treatment-naive tumors or emerge in response to therapy. The combination of an AXL-MMAE antibody-drug conjugate with BRAF and MEK inhibitors eliminates heterogeneous melanoma cell populations and prolongs survival in experimental in vivo models at tolerable toxicity. This approach is currently being tested in clinical trials and provides insights into the therapeutic targeting of intra-tumor heterogeneity.
