Single-cell RNA sequencing illuminates the ontogeny, conservation and diversification of cartilaginous and bony fish lymphocytes

Both cartilaginous and bony fishes have adaptive immune cells, but the evolutionary features of these diverse cell populations remain understudied. Here the authors report single-cell RNA sequencing atlas of both species to define these features and to find reduced evolutionary diversification of immune cells in cartilaginous fishes.

Discovery of an unconventional lamprey lymphocyte lineage highlights divergent features in vertebrate adaptive immune system evolution

Lymphocyte subsets remain inadequately understood in jawless vertebrates, relative to jawed vertebrates. In this study, the authors combine single-cell RNA and whole-transcriptome sequencing to demonstrate that the lamprey, Lampreta morii, harbours specialised T-like cells that are distinct from those found in jawed vertebrates.

Double Asteroid Redirection Test (DART) mission’s impact on asteroid Dimorphos has led to various impact related features. Here, the authors show that those features result naturally from the dynamical interaction of the ejecta with the binary system and solar radiation pressure.

Genome-wide association analyses identify 143 risk variants and putative regulatory mechanisms for type 2 diabetes

GWAS have so far identified 129 loci associated with type 2 diabetes (T2D). Here, the authors meta-analyse three large T2D GWA studies which uncovers 42 additional loci, further prioritize 33 functional genes using eQTL and mQTL data and propose regulatory mechanisms for three putative T2D genes.

The geology and evolution of the Near-Earth binary asteroid system (65803) Didymos

Images collected during NASA’s DART mission of the asteroid Didymos and its moon, Dimorphos, are used to explore the origin and evolution of the binary system. Authors analysis indicate that both asteroids are weak rubble piles and that Didymos’ surface should be about 40 to 130 times older than Dimorphos.

Integration of summary data from GWAS and eQTL studies predicts complex trait gene targets

Jian Yang and colleagues propose a method that integrates summary data from GWAS and eQTL studies to identify genes whose expression levels are associated with complex traits because of pleiotropy. They apply the method to five human complex traits and prioritize 126 genes for future functional studies.

Global genetic differentiation of complex traits shaped by natural selection in humans

Human complex traits and disorders often show differentiation among populations. Here, the authors analyze SNPs associated with 10 complex traits using data of large sample size from African, East Asian and European populations, and find significant genetic differentiation among global populations shaped by natural selection.

Genetic control of RNA splicing and its distinct role in complex trait variation

A powerful method for splicing quantitative trait loci (sQTL) mapping, THISTLE, is presented and applied to a collection of 2,865 brain samples. Integration with GWAS identifies 244 genes associated via cis-sQTLs, of which 61% were not identified using expression QTLs.

Identifying gene targets for brain-related traits using transcriptomic and methylomic data from blood

To comprehend the genetic regulatory mechanisms underlying brain-related traits in humans, Qi et al. estimate the correlation of expression and DNA methylation QTL effects in cis between blood and brain and show that using blood eQTL/mQTL data of large sample size can increase power in gene discovery for brain-related traits and diseases.

Causal associations between risk factors and common diseases inferred from GWAS summary data

Genetic methods are useful to test whether risk factors are causal for or consequence of disease. Integrative analysis of omics summary data reveals putative mechanisms underlying complex traits

The identification of the causal gene at a GWAS locus remains to be a challenging task. Here, using the SMR & HEIDI method to integrate GWAS, eQTL and mQTL data, Wu et al. map DNA methylation sites to the transcriptome and thereby prioritize functionally relevant genes for 12 human complex traits.
