LRIG1 is a pleiotropic androgen receptor-regulated feedback tumor suppressor in prostate cancer

LRIG1 is a tumour suppressor in several cancers. Here, the authors show that androgen receptor directly transactivates LRIG1 which then antagonises ERBB signalling and c-Myc expression to inhibit prostate tumorigenesis.

Obesity increases genomic instability at DNA repeat-mediated endogenous mutation hotspots

Obesity is associated with cancer, though the mechanistic links are unclear. Here, the authors find that obesity increased genetic instability induced by DNA repeat sequences and reduced DNA repair in mice. This study provides mechanistic insight into the obesity-cancer link.

Aerobic glycolysis and diminished oxidative phosphorylation exhibited by tumour cells enables the production of energy necessary to support malignant proliferation. Here the authors show that UCP3 promotes mitochondrial uncoupling and prevents tumorigenesis through a mitochondrially-driven pathway of AKT inhibition.

Obesity and prostate cancer — microenvironmental roles of adipose tissue

Current evidence suggests that adipose stromal cells, a component of peri-prostatic white adipose tissue and the tumour microenvironment, have an important role in driving aggressive prostate cancer in obesity. These cells are potential targets of therapies to suppress cancer aggressiveness in obesity.
