SETD1A modulates cell cycle progression through a miRNA network that regulates p53 target genes

The p53-inducible antiproliferative gene BTG2 is suppressed in many cancers, in the absence of inactivating gene mutations. Here the authors show that the histone lysine methyltransferase SETD1A suppresses the expression of several p53 target genes including BTG2by inducing a network of microRNAs.

Two JmjC-domain-contain ing enzymes are shown to catalyse demethylation of histone H3K27me3, a histone modification mediated by polycomb proteins and associated with stem cell maintenance and differentiation.

DNA replication fork speed underlies cell fate changes and promotes reprogramming

Totipotent cells in mouse embryos and 2-cell-like cells have slow DNA replication fork speed. Perturbations that slow replication fork speed promote 2-cell-like cell emergence and improve somatic cell nuclear transfer reprogramming and formation of induced pluripotent stem cell colonies.

Genome-wide identification of microRNAs regulating cholesterol and triglyceride homeostasis

From a systematic analysis of genome-wide association studies of blood lipid levels, Wagschal et al. identify several miRNAs that target key proteins involved in cholesterol and lipid metabolism, including the LDL receptor and the ABCA1 cholesterol transporter.

Histone H3 methylation links DNA damage detection to activation of the tumour suppressor Tip60

Tip60 acetylation of ATM kinase is necessary for DNA double-strand break repair and cancer suppression. Tip60 is recruited to breaks by histoneH3 trimethylated on lysine 9 and the Mre11/Rad50/Nbs1 damage response complex, activating its acetyltransferase activity.
