RNA catalyses nuclear pre-mRNA splicing

The spliceosome is shown to catalyse splicing through the RNA and not the protein components of the spliceosome; pre-messenger RNA splicing requires U6 snRNA acting by a mechanism similar to that used by group II self-splicing introns.

Evidence for a group II intron–like catalytic triplex in the spliceosome

It has long been thought that the catalytic RNAs of self-splicing group II introns and the spliceosome function by similar mechanisms. Now, a combination of genetic, cross-linking, and biochemical analyses of yeast U6 snRNA provide compelling evidence that spliceosomal RNAs form triplex structures similar to those used by group II introns to catalyze splicing.

The debranching enzyme Dbr1 regulates lariat turnover and intron splicing

Dbr1 exhibits debranching specificity and effect on splicing. Here the authors combine co-immunoprecipitation, RNA binding and lariat analysis and suggest a role for Dbr1 interactor AQR in intron recycling. Dbr1 depletion leads to increased dwell time of spliceosome on excised lariats.

Global impact of unproductive splicing on human gene expression

A conformational switch in PRP8 mediates metal ion coordination that promotes pre-mRNA exon ligation

The core of the spliceosome contains PRP8, a highly conserved protein that contacts each of the active site components and is implicated in both steps of pre-mRNA splicing. New work shows that structural rearrangement of the PRP8 RNase H domain between the first and second steps forms a Mg²⁺-binding site that is required for the second splicing reaction, revealing the molecular basis of the two-state model of splicing catalysis.

Reverse transcriptases lend a hand in splicing catalysis

The first high-resolution views of group II intron maturases illuminate the architectural and functional roles of these multidomain proteins in splicing and DNA invasion. The maturases show striking structural and functional homology to a central protein involved in spliceosomal pre–messenger RNA splicing, thus reinforcing the idea that group II introns and the spliceosome descended from a common ancestor.
