Normothermic ex vivo kidney perfusion preserves mitochondrial and graft function after warm ischemia and is further enhanced by AP39

The authors previously reported that normothermic ex vivo kidney perfusion is superior to static cold storage in terms of organ protection, but the detailed mechanism was unclear. Here the authors show that the organ-protective effects of normothermic perfusion are mediated by maintenance of mitochondrial function and enhanced by administration of AP39, a mitochondria-targeted hydrogen sulfide donor.

Coordinated inflammatory responses dictate Marburg virus control by reservoir bats

The authors show pro-inflammatory responses are needed for Marburg virus control in its natural bat reservoir, and that if reduced, humanlike disease and shedding results, suggesting that natural immunomodulatory stressors may increase spillover risk.

Defective Wnt-dependent cerebellar midline fusion in a mouse model of Joubert syndrome

Joubert syndrome is a developmental disease affecting multiple parts of the body, including the cerebellum in the brain. Now, Joseph Gleeson and his colleagues show that mouse models of Joubert syndrome show deficient Wnt-dependent cerebellar proliferation and midline fusion and that these phenotypes can be rescued by lithium, a Wnt agonist.

Subcellular spatial regulation of canonical Wnt signalling at the primary cilium

Through the action of intraflagella transport, primary cilia are found to sequester Jouberin, a positive regulator of Wnt signalling, to limit β-catenin nuclear entry and to modulate Wnt responsiveness. Consequently, Wnt is exacerbated in cells without cilia but inhibited in cells with more than one cilium.

Impaired Wnt–β-catenin signaling disrupts adult renal homeostasis and leads to cystic kidney ciliopathy

Canonical Wnt signaling is known to be crucial in embryonic organ development. Joseph Gleeson and his colleagues now report that it is also important in the adult homeostasis of the kidney, especially after injury, and that disruption of this signaling pathway results in cystic kidney disease.
