Direct control of regulatory T cells by keratinocytes

Skin is constantly exposed to environmental stressors. Georgopoulos and colleagues show that regulatory T cells respond to the cytokine TSLP produced by stressed keratinocytes and that a loss of skin T_reg cell expression of TSLPR leads to lethal inflammation.

Harnessing of the nucleosome-remodeling-deacetylase complex controls lymphocyte development and prevents leukemogenesis

Lymphocyte development is regulated by Ikaros transcription factors. Georgopoulos and colleagues show that Ikaros tethers the nucleosome-remodeling factor Mi-2β and restrains its ability to act at nonlymphoid gene sites.

A double-negative thymocyte-specific enhancer augments Notch1 signaling to direct early T cell progenitor expansion, lineage restriction and β-selection

Notch signaling is required for T cell development. Georgopoulos and colleagues identify an enhancer that specifically boosts Notch1 expression in early thymic progenitors through the DN3 stage, expanding the less committed multipotent progenitors and preparing these cells for faithful lineage commitment.

GATA-3 controls self-renewal in stressed HSCs

The transcription factor GATA-3 is expressed in quiescent hematopoietic stem cells (HSCs) with long-term repopulating ability. Stress-mediated activation of HSCs promotes localization of GATA-3 to the nucleus in a manner dependent on the mitogen-activated protein kinase p38α and restricts self-renewal.

STAG2 regulates interferon signaling in melanoma via enhancer loop reprogramming

STAG2 is a core subunit of the cohesin complex involved in DNA looping, but its transcriptional targets are largely unknown. Here the authors show STAG2 controls the 3D chromatin structure at the IRF9 locus to restrict IRF9 expression. Loss of STAG2 results in IRF9 activation, which in turn upregulates PD-L1 expression in cancer cells, suggesting a tumor suppressor function in immune evasion.

How signals through the pre–B cell antigen receptor (pre-BCR) and IL-7 receptor (IL-7R) coordinate population expansion of pre-B cells with subsequent recombination of the immunoglobulin κ-chain locus is unclear. Clark and colleagues show that pre-BCR signaling via the Ras-MEK-Erk pathway poises pre–B cells to undergo differentiation after escaping IL-7R signaling.
