Are we lowering LDL cholesterol sufficiently?

A huge body of evidence now supports aggressive LDL cholesterol lowering to reduce cardiovascular risk, but what LDL cholesterol target level should be recommended? Here, Philip Barter and Kerry-Anne Rye examine the safety issues associated with achieving very low levels of LDL cholesterol and whether it is feasible to achieve very low levels in all high-risk people.

Scavenger receptor class B1 — a target to reduce CHD risk?

A loss-of-function mutation in the gene encoding scavenger receptor class B member 1 (SRB1) has been identified. This mutation increases the risk of coronary heart disease (CHD) despite being associated with increased concentration of HDL cholesterol. Upregulation of SRB1 expression might be a novel therapeutic approach for reducing CHD risk.

HDL-transferred microRNA-223 regulates ICAM-1 expression in endothelial cells

Lipoproteins such as HDL can bind and transport microRNAs throughout the body. Here the authors provide a new mechanism contributing to the anti-inflammatory effects of HDL by which HDL-associated miR-223 is transferred to endothelial cells, where it inhibits expression of the adhesion molecule ICAM-1.

Optimized negative-staining and cryo–positive staining EM reveals that human cholesteryl ester transfer protein penetrates into HDL and LDL from each distal end and potentially forms a continuous tunnel by connecting its internal series of isolated hydrophobic cavities together for cholesteryl ester transfer.
