HS1BP3 negatively regulates autophagy by modulation of phosphatidic acid levels

Autophagy must be tightly controlled at each step of the process. Here the authors show that HS1BP3 binds phosphatidic acid (PA) at autophagosome precursors and negatively regulates autophagosome formation by modulating the activity and localization of the PA-producing enzyme phospholipase D1.

ANCHR mediates Aurora-B-dependent abscission checkpoint control through retention of VPS4

In the presence of incorrectly segregated chromosomes, the Aurora-B-dependent abscission checkpoint prevents the final stage of cytokinesis. Stenmark and colleagues identify a role for the previously uncharacterized protein ANCHR at the checkpoint, where it acts in an Aurora-B-dependent manner to retain the ATPase VPS4 at the midbody.

PtdIns(3)P controls cytokinesis through KIF13A-mediated recruitment of FYVE-CENT to the midbody

A new link is reported between regulators of endosomal trafficking and cytokinesis. Production of the phosphoinositide lipid PI3P at the midbody triggers the KIF13A-mediated recruitment of the centrosomal proteins FYVE-CENT and TTC19 to the division site. TTC19 may in turn regulate abscission by control of the ESCRTIII complex.

DNA copy number motifs are strong and independent predictors of survival in breast cancer

Pladsen et al. develop Copy Aberration Regional Mapping Analysis (CARMA), an algorithm that derives motifs for copy number profiles in breast cancers by integrating several features, to predict breast cancer prognosis and stratifications. Their algorithm can detect replication and repair defects and can be used in personalized medicine.
