Intratumoral delivery of FLT3L with CXCR3/CCR5 ligands promotes XCR1⁺ cDC1 infiltration and activates anti-tumor immunity

XCR1+ type 1 conventional DCs (cDC1s) are crucial to mount anti-tumor immune responses, however their infiltration within tumours is often limited. Here the authors show that cDC1 infiltration could be expanded by intratumoral delivery of mesenchymal stromal cells engineered to express the membrane bound form of FLT3L in combination with poly(I:C) or CXCL9 and CCL5, improving anti-tumor immunity in preclinical models.

IRAP⁺ endosomes restrict TLR9 activation and signaling

The receptor TLR9 needs to be carefully regulated to avoid recognition of self nucleic acids and ensuing autoinflammation. Saveanu and colleagues demonstrate a further level of regulation through the retention of TLR9 in IRAP⁺ endosomes.

IRAP-dependent endosomal T cell receptor signalling is essential for T cell responses

T cell receptors (TCR) are internalized when activated by their ligands. Here the authors show that the internalized TCRs are localized to endosomes expressing IRAP and Syntaxin 6 to maintain intracellular signalling capacity, whose importance is shown by the absence of efficient polyclonal anti-tumour response in mice with T-specific conditional deletion of IRAP.

Defective autophagy in CD4 T cells drives liver fibrosis via type 3 inflammation

Liver fibrosis is a consequence of the sustained inflammatory processes underpinning chronic liver disease. Here authors show that autophagy in CD4 T cells is an important process in preventing the emergence of pathogenic Th17 cells, which are causal to the progression of liver fibrosis.

PD-L1- and IL-4-expressing basophils promote pathogenic accumulation of T follicular helper cells in lupus

Basophils have been implicated in systemic lupus erythematosus (SLE), as evidenced by the fact that basophil-deficient mice do not develop the disease. Here, the authors demonstrate that PD-L1 and IL-4 expression in basophils promotes the pathogenic accumulation of follicular helper T cells in patients with SLE and murine models.
