Front-line therapy in lung cancer with mutations in EGFR

Large randomized phase III trials conducted in patients with non-small-cell lung cancer (NSCLC) harboring activating mutations in EGFR have demonstrated that erlotinib or gefitinib are superior to platinum-based chemotherapy. Zhou et al. have now confirmed that these agents represent the best treatment we can offer today as front-line therapy for EGFR-mutant NSCLC.boxed-text

Rare missense variants in POT1 predispose to familial cutaneous malignant melanoma

Maria Teresa Landi and colleagues identify a rare missense variant in POT1 shared by five melanoma-prone families from Italy and associated with increased telomere length and telomere fragility. They also identify additional familial melanoma cases with rare missense variants in POT1 and find a significant excess of rare exonic POT1 variants in melanoma cases compared to controls, implicating POT1 variants in melanoma susceptibility.

Genome-wide association meta-analyses combining multiple risk phenotypes provide insights into the genetic architecture of cutaneous melanoma susceptibility

Meta-analysis of 36,760 cases and 375,188 controls identifies 54 loci associated with susceptibility to cutaneous melanoma. Further analysis combining nevus count and hair color GWAS results provide insights into the genetic architecture of melanoma.

Circulating tumour DNA in early stage and locally advanced NSCLC: ready for clinical implementation?

Analysis of circulating tumour DNA (ctDNA) is commonly used for molecular profiling in patients with advanced-stage non-small-cell lung cancer (NSCLC). The authors of this Review summarize the available evidence on the potential utility of incorporating ctDNA in the management of those with early stage and locally advanced NSCLC and propose interventional studies to provide the necessary additional evidence.
