A BCL-xL/BCL-2 PROTAC effectively clears senescent cells in the liver and reduces MASH-driven hepatocellular carcinoma in mice

Yang, Jn-Simon, He et al. report that the dual BCL-xL/BCL-2 PROTAC 753b is a potent and liver-tropic senolytic, which (unlike other inhibitors of BCL-xL) does not cause severe thrombocytopenia. They evaluate its efficacy in natural aging and in reducing progression from steatotic liver disease to hepatocellular carcinoma, using mouse models.

Reconstitution of human PDAC using primary cells reveals oncogenic transcriptomic features at tumor onset

The cellular origin of human pancreatic ductal adenocarcinoma (PDAC) is still unclear. Here the authors used human primary acinar and ductal cells to successfully reconstitute PDAC tumorigenesis from both lineages and revealed transcriptional changes during early PDAC progression.

Attenuation of RNA polymerase II pausing mitigates BRCA1-associated R-loop accumulation and tumorigenesis

The vast majority of BRCA1-driven breast cancers derive from luminal progenitor cells but the mechanisms of this lineage specificity are unclear. Here the authors show that dangerous accumulation of DNA-RNA hybrid structures due to RNA polymerase II pausing, occurs specifically in luminal epithelial cells.

Differential effects of GLI2 and GLI3 in regulating cervical cancer malignancy in vitro and in vivo

GLI transcription factors, which mediate the Hedgehog pathway, are over-expressed in cervical cancer. RNA interference reveals an essential role of GLI2, but not GLI3, in promoting proliferation and migration and xenograft tumor growth of human cervical cancer cells. Mechanistically, the function of GLI2 is mediated by protein kinase B (AKT), and may be targeted as a novel therapeutic strategy.
