Crocodile defensin (CpoBD13) antifungal activity via pH-dependent phospholipid targeting and membrane disruption

Defensins are a class of host defence peptides that contribute to the immune system of eukaryotes. Here, the authors report the structure of saltwater crocodile defensin CpoBD13 and the mechanism of pH-dependent antifungal activity.

X-ray structure of a carpet-like antimicrobial defensin–phospholipid membrane disruption complex

Defensins are cationic antimicrobial peptides that permeabilize membranes of pathogens presumably via the assembly of defensin–lipid oligomers. Here authors provide evidence for this by solving the crystal structure of a plant defensin in an oligomeric state with phospholipids.

A broadly cross-reactive i-body to AMA1 potently inhibits blood and liver stages of Plasmodium parasites

Apical membrane antigen-1 (AMA1) is important for Plasmodium parasite invasion, but surface polymorphisms in P. falciparum AMA1 hamper vaccine and antibody development. Here the authors identify a humanised single-domain antibody molecule, an i-body, that binds to AMA1 of all Plasmodium parasites examined and inhibits parasite invasion.

Structural basis of coronavirus E protein interactions with human PALS1 PDZ domain

Airah Javorsky et al. present the crystal structures of SARS-CoV-1 and SARS-CoV-2 E proteins in complex with the PALS1 PDZ domain. Their results suggest that the coronavirus E protein can interfere with normal PALS1 binding, potentially disrupting epithelial tissue integrity, and may provide future insight into the development of small molecule inhibitors against SARS-CoV-1/2.
