Somatic CRISPR/Cas9-mediated tumour suppressor disruption enables versatile brain tumour modelling

Gene transfer is a powerful technique to investigate the mechanistic basis of tumorigenesis. Here Zuckermann et al. adapt CRISPR/Cas9 genome editing to target potential oncogenes somatically in vivo, establishing a fast and convenient system for validating novel genetic candidates.

Recurrent somatic alterations of FGFR1 and NTRK2 in pilocytic astrocytoma

Stefan Pfister and the ICGC PedBrain Tumor Project report whole-genome sequencing of 96 pilocytic astrocytomas. They identify recurrent activating mutations in FGFR1 and PTPN11 and novel NTRK2 fusion genes.

YAP1 subgroup supratentorial ependymoma requires TEAD and nuclear factor I-mediated transcriptional programmes for tumorigenesis

The molecular mechanisms driving proliferation in the pediatric brain cancer epdendymoma are poorly understood. Here the authors show that a YAP1- MAMLD1 fusion drives tumor formation in mice and show that the fusion protein can collaborate with the TEAD and NFI transcription factors.

Chd7 is indispensable for mammalian brain development through activation of a neuronal differentiation programme

Mutations in the chromatin modifier Chd7 have been associated with CHARGE syndrome and other developmental disorders. Here the authors show that Chd7 is required for the activation of genes essential for cerebellar granule cell differentiation, and that disrupting Chd7 leads to cerebellar hypoplasia in mice.
