Hexameric NuMA:LGN structures promote multivalent interactions required for planar epithelial divisions

LGN and NuMA link epithelial polarity sites with astral microtubules and associate with dynein, but the architecture of such cortical force-generating complexes is unknown. Here, the authors report the crystal structure of NuMA:LGN hetero-hexamers, and unveil their role in promoting the assembly of active cortical dynein/dynactin motors.

Localized Wnt-signaling promotes asymmetric NuMA-dependent oriented divisions and unequal apportioning of mitochondria

Asymmetric cell division often requires alignment of the mitotic spindle to cortical polarity cues. Here the authors show that cortical Wnt signaling induces formation of a complex between NuMA/dynein microtubule motors, LRP6 and β-catenin that promotes asymmetric division.

Drosophila TNFRs Grindelwald and Wengen bind Eiger with different affinities and promote distinct cellular functions

The Drosophila tumour necrosis factor (TNF) system comprises a single ligand Eiger (Egr) and two receptors. The structure of Egr in complex with the extracellular domain of the receptor Grindelwald and accompanying data suggest that distinct affinities of TNF ligand for its receptors mediate non-redundant functions.

Recognition and inhibition of SARS-CoV-2 by humoral innate immunity pattern recognition molecules

Stravalaci et al. examined recognition of SARS-CoV-2 by human soluble innate pattern recognition receptor. They report that pentraxin 3 and mannose-binding protein recognize viral nucleoprotein and spike, respectively. Mannose-binding lectin has antiviral activity, and human genetic polymorphisms of MBL2 are associated with more severe COVID-19.

Symmetric dimethylation of H3R2 is a newly identified histone mark that supports euchromatin maintenance

Although the asymmetric dimethylation of histone H3R2 acts as a repressive mark, new studies reveal that symmetrically dimethylated H3R2 (H3R2me2s) is a functional histone mark in vivo. The RBBP7 co-repressor is excluded from binding H3R2me2s in favor of the coactivator WDR5, which poises euchromatic genes for transcription activation upon cell-cycle exit and differentiation.
