Through sedimentation velocity-based methods, Igel-Egalon et al. characterize the mechanism of prion protein misfolding during early prion replication. They identify two subsets of structurally different assemblies, Ai and Bi, and two sequential processes, the first generating Ai and the second transforming Ai into Bi.
