Short single-stranded DNA degradation products augment the activation of Toll-like receptor 9

DNA degradation products are frequently found in the endosome, but how they regulate the activation of Toll-like receptors is not known. Here the authors show that single-stranded DNA as short as two nucleotides can enhance the ability of longer DNA oligonucleotides to activate Toll-like receptors.

A second binding site for double-stranded RNA in TLR3 and consequences for interferon activation

Toll-like receptor 3 (TLR3) recognizes double-stranded RNA (dsRNA) molecules produced by many viruses and activates an inflammatory response. Synthetic dsRNAs such as small interfering RNAs have been shown to activate TLR3. Now the TLR3 ectodomain is found to contain two dsRNA binding sites, and the implications for dsRNA recognition and selectivity and downstream signaling are discussed.

Cytokine-armed pyroptosis induces antitumor immunity against diverse types of tumors

‘Gasdermin-mediated pyroptosis can induce anti-tumor immune responses. Here the authors report a toolbox of gasdermin D variants for induction of pyroptosis, showing that, when combined with intratumoral expression of cytokines (IL-1β, IL-12, or IL-18), anti-tumor immune responses in preclinical models are enhanced.
