STIM1-mediated gating of CRAC channels occurs through a mechanism in which ion selectivity and gating are closely coupled, and the residue V102 is identified as a candidate for the channel gate.

STIM1 activates CRAC channels through rotation of the pore helix to open a hydrophobic gate

Store-operated Ca²⁺channels (CRAC) are involved in several cellular functions. Here the authors identify a hydrophobic gate in the CRAC pore and show that CRAC activation by STIM1 involves rotation of the pore helix that hydrates this region to allow ion passage through the pore.

Conformational dynamics of STIM1 activation

The binding of stromal interaction molecule 1 (STIM1) to ORAI calcium channels is critical for store-operated calcium entry (SOCE), a calcium influx pathway conserved among nearly all vertebrate cells. Although many major steps of this pathway are well understood, crucial details regarding the mechanism of STIM1 activation remain unclear. A study in this issue provides important new insights into the conformational changes that occur during STIM1 activation.

Cavβ1 regulates T cell expansion and apoptosis independently of voltage-gated Ca²⁺ channel function

The function of voltage-gated calcium channels in T cells is not well understood and controversial. Here the authors report that a regulatory beta subunit of voltage-gated calcium channels regulates T cell function despite no evidence that these channels were functional within T cells during activation.
