Despite the implied allosteric modulation of the M. tuberculosis 20S core particle (CP), the specific conformations involved remained elusive. Here, the authors use cryo-EM and HDX-MS to determine the 20S CP structure in an auto-inhibited state that is distinguished from the canonical resting state.
