  • Acid-sensing ion channels (ASICs) are crucial in pain sensation linked to extracellular acidification, yet selective modulators for ASIC3 remain limited. Here, the authors identify WRPRFa, a selective and potent peptide modulator of ASIC3 gating, that enhances ASIC3 pH sensitivity and removes acute desensitization, offering a valuable tool for studying ASIC3 gating mechanisms.

  • In voltage-gated potassium (KV) channels, the voltage-sensing domain (VSD) undergoes activation states to trigger pore opening via electro–mechanical (E–M) coupling. Here authors show that KV7.1 undergoes a two-stage E–M coupling mechanism during voltage-dependent activation.

  • Yongfeng Liu, Xianjin Xu, Junyuan Gao et al. perform in silico screening targeting compounds that resemble both KCNQ1 and PIP2 structures and identify CP1 as a compound that could restore KCNQ1 currents after PIP2 depletion. Like PIP2, this compound can mediate VSD-pore coupling of the channel, albeit via a different mechanism from PIP2.
