Cytokinetic-based rings for self-division of synthetic cells lack spatial control for the targeting of contractile forces within cell-like lipid vesicles. Here, authors combine actomyosin rings and the MinDE bacterial protein system to achieve ring positioning, equatorial furrowing and blebbing.
