Interleukin (IL)-1α is a critical inflammatory cytokine and yet its intracellular role is poorly understood. Here, the authors use proximity labelling of pro-IL-1α and assess its interactome, and dissect the presence and impact on a conserved nuclear interaction and its impact on the immunobiology and evolutionary convergence/divergence between species.
