Structures of mouse and human GITR–GITRL complexes reveal unique TNF superfamily interactions

Glucocorticoid-induced tumor necrosis factor receptor-related protein (GITR) and GITR ligand (GITRL) regulate immune cell activities, including anti-tumor immune responses. Structures and visualization of human and mouse GITR–GITRL complexes offer insight into the architecture of higher-order membrane assemblies, and their signaling.

Molecular mechanism of the synaptotagmin–SNARE interaction in Ca²⁺-triggered vesicle fusion

The interaction between synaptotagmin and SNAREs was characterized by a combination of single-molecule FRET and crystallography. The arrangement of the two Ca²⁺-binding loops of synaptotagmin 3 within SNARE-induced Ca²⁺-bound synaptotagmin 3 matches that of SNARE-bound synaptotagmin 1, suggesting a common molecular mechanism by which the synaptotagmin–SNARE interaction plays a role in Ca²⁺-triggered vesicle fusion.

An autonomous CDR3δ is sufficient for recognition of the nonclassical MHC class I molecules T10 and T22 by γδ T cells

The molecular basis of ligand recognition by γδ TCRs remains vague. Adams and colleagues show that the CDR3δ loop, when grafted in place of a CDR3α loop, is sufficient to confer γδ TCR specificity on an αβ TCR.

The role of MscL amphipathic N terminus indicates a blueprint for bilayer-mediated gating of mechanosensitive channels

The activation of bacterial mechanosensitive channels is still not fully understood. Here, Bavi et al. show that the N-terminal helix of MscL dynamically couples membrane tension to channel gating, suggesting a conserved mechanism underlying the mechanosensitivity of ion channels of higher organisms.

Single-molecule FRET–derived model of the synaptotagmin 1–SNARE fusion complex

Single-molecule FRET studies have resulted in an experimentally derived model of a synaptotagmin–SNARE complex. In this complex of SNARE with synaptotagmin 1, the arrangement of the Ca²⁺-binding loops is similar to that of the structure of SNARE-induced Ca²⁺-bound synaptotagmin 3. This suggests a common molecular mechanism by which the synaptotagmin–SNARE interaction plays a role in Ca²⁺-triggered vesicle fusion.
