Organoids in endocrine and metabolic research: current and emerging applications

Organoid technologies are a potent tool for investigating human biology, modelling diseases and developing novel therapies. In this Viewpoint, experts in metabolic and endocrine research in the brain, pituitary, skeletal muscle, bone and gastrointestinal system discuss how organoids and related bioengineered systems are currently used in their field and how innovations in these technologies could transform future research.

Rejuvenation of the muscle stem cell population restores strength to injured aged muscles

In two new studies, Helen Blau and Bradley Olwin and their colleagues show that muscle stem cells in aged mice have an intrinsic defect in their stem cell capacity, contributing to age-related sarcopenia. They also provide mechanistic insight to explain this phenomenon and show that biochemical and biophysical manipulations can overcome the defect, suggesting a possible future route of therapy.

TGFβ signaling curbs cell fusion and muscle regeneration

The fusion of muscle progenitor cells to form syncytial myofibers is required for skeletal muscle development and regeneration. Here, the authors describe a novel and specific molecular regulation of muscle cell fusion driven by transforming growth factor beta (TGFβ) signaling.

Tissue mechanics modulate microRNA-dependent PTEN expression to regulate malignant progression

Mouw et al. delineate a molecular pathway by which matrix stiffness promotes tumor malignancy. Upon matrix stiffening, integrin signaling is engaged, triggering a signaling cascade that regulates the protumorigenic microRNA miR-18a. This leads to downregulation of PTEN and activation of oncogenic signaling. The pathway provides a link between mechanotransduction, miR regulation and oncogene activation that integrates biophysical changes into tumor progression and can also be altered in human tumors.
