TERRA transcription destabilizes telomere integrity to initiate break-induced replication in human ALT cells

TERRA RNA has previously been linked to Alternative lengthening of telomeres (ALT). Here the authors developed a tool to rapidly inhibit TERRA transcription from different chromosome ends in an ALT cell line to show that TERRA transcription actively promotes break induced replication (BIR) and destabilizes telomere integrity in ALT cells.

RNaseH1 regulates TERRA-telomeric DNA hybrids and telomere maintenance in ALT tumour cells

A subset of cancers maintains telomere length independently of telomerase by activating alternative lengthening of telomeres (ALT) pathways. Here the authors show that RNaseH1 modulates telomeric homologous recombination frequencies in ALT cells by regulating the levels of RNA–DNA hybrids between TERRA and telomeric DNA.

FANCM limits ALT activity by restricting telomeric replication stress induced by deregulated BLM and R-loops

In cancer cells, telomeres can be elongated through homology directed-repair pathways in a process known as Alternative Lengthening of Telomeres (ALT). Here, the authors reveal that FANCM regulates ALT activity and ALT cell proliferation by limiting the activity of uncontrolled BLM and telomeric R-loops.

TRF1 participates in chromosome end protection by averting TRF2-dependent telomeric R loops

In vitro and cellular assays unexpectedly reveal that shelterin protein TRF2 binds TERRA and stimulates strand invasion within telomere repeats and that TRF1 suppresses this activity to prevent telomere loss and genome instability.
