Prospective evaluation of genomics-guided off-label treatment

Evaluation by the Drug Rediscovery Protocol of off-label use of 37 approved cancer drugs on 1,610 patients showed modest activities overall, although subgroups of responders suggest that off-label drug use could be beneficial in tandem with more stringent biomarker selection.

Atheroprotective communication between endothelial cells and smooth muscle cells through miRNAs

Dimmeler and colleagues show that the atheroprotective transcription factor KLF2 activates expression of the microRNAs miR-143/145 in endothelial cells. miR-143/145 are subsequently enriched in secreted microvesicles and taken up by smooth muscle cells to elicit anti-atherogenic responses.

MicroRNA-34a regulates cardiac ageing and function

A role is demonstrated for miR-34a, a microRNA that is upregulated in the ageing heart; miR-34a downregulates PNUTS, a protein that protects cardiomyocytes and telomeres, silencing of miR-34a is therefore a promising therapeutic target.

MicroRNAs in myocardial infarction

MicroRNAs regulate the processes that underlie cardiac remodelling after myocardial infarction, including angiogenesis and cardiomyocyte apoptosis and proliferation. In this Review, Boon and Dimmeler summarize the microRNAs that are involved with these postischaemic pathways, and discuss potential therapeutic interventions to enhance cardiac regeneration in patients with acute myocardial infarction.

Cardiomyocytes stimulate angiogenesis after ischemic injury in a ZEB2-dependent manner

ZEB2 transcription factor is increased in a subset of cardiomyocytes during stress to induce cardioprotective effects after injury. Here the authors show that therapeutic delivery of ZEB2 prevents cardiac dysfunction after ischemic damage and promotes the activation of pro-angiogenic signals.

Winski et al. evaluate the miRNA abundance in plasma of around 200 patients with abdominal aortic aneurysm (AAA) and matched controls. MiR-15a is identified as a candidate biomarker and AAA mouse models and additional patient samples are used to determine miR-15a’s localization, expression, and the impact of miR-15a dysregulation.

Long non-coding RNA LASSIE regulates shear stress sensing and endothelial barrier function

Stanicek et al identify a shear stress-induced long non-coding RNA they name LASSIE, which stabilises junctions between endothelial cells through interactions with junctional and cytoskeletal proteins. This study provides insights into how a transcript that does not encode a protein controls endothelial response to forces associated with blood flow and endothelial barrier function.
