Drug-coated balloon therapy in coronary and peripheral artery disease

De novostenosis of coronary or peripheral arteries as well as restenosis of previously stented vessels are increasingly prevalent clinical problems. In this Review, Byrne and colleagues summarize the evidence for the use of novel balloon catheters covered with antiproliferative drug both to restore vessel patency and to provide long-lasting inhibition of cell regrowth.

Targeted sampling of natural product space to identify bioactive natural product-like polyketide macrolides

Polyketide macrolides are of interest for drug discovery but their inherent structural and stereochemical complexity hinders the exploration of related regions of chemical space more broadly. Here, the authors designed in silico and synthesized a library of tetrahydrofuran-containing polyketide macrolides, and screened them against a panel of biological assays, identifying biologically active library members.

Total synthesis of nine longiborneol sesquiterpenoids using a functionalized camphor strategy

Generating strategies that use modern synthetic methods to access privileged chemical space is a central goal of total synthesis. Now, a strategy that is orthogonal to classic approaches, coupled with C–H functionalization methods, leads to the shortest synthesis of the sesquiterpenoid longiborneol and divergent syntheses of oxygenated longiborneol congeners.

Organosodium compounds are appealing sustainable alternatives to precious transition metal compounds, yet they have rarely been used in homogeneous catalysis. Here, the authors introduce a dihydropyridylsodium compound, Na1,2-tBu-DH(DMAP), and its monomeric variant [Na-1,2-tBuDH(DMAP)]·Me₆TREN, and demonstrate their effectiveness in transfer hydrogenation catalysis of a representative alkene, 1,1-diphenylethylene, using 1,4-cyclohexadiene as a hydrogen source.

Protease inhibitors represent attractive antiviral agents against coronaviruses, however, current inhibitors show restricted efficiency which limits their use. Here, the authors develop covalent reversible peptidyl inhibitors with nitroalkene warheads, showing inhibition against main protease and human Cathepsin L, preventing viral infection in a cellular assay.

Inhibition of Delta-induced Notch signaling using fucose analogs

Protein O-fucosyltransferase 1 (Pofut1) regulates Notch activity by adding O-fucose residues to its extracellular domain. Fucose analogs were identified that inhibited Delta-mediated Notch binding and activation but spared Jagged1-mediated signaling.
