  • Repeat-rich sequences play a role in 3D genome architecture in higher eukaryotes; however, this remains unexplored in brain cells. Here, the authors show that upregulation of endogenous retroviral (ERV) sequences is linked to changes in the 3D structure in the brain, which is also observed by comparison of mouse strains with recent retrotransposon expansion.

  • Isolation during critical periods of development prevents development of normal social behaviours in mice, and this is thought to involve the prefrontal cortex. Here, the authors identify an activation pattern in parvalbumin-positive interneurons in the dorsal medial prefrontal cortex that when activated promotes sociability behaviours in mice.

  • Schahram Akbarian and colleagues report that mutation of the gene encoding the SETDB1 (KMT1E) histone methyltransferase in mouse neurons leads to dissolution of chromosome conformations and a topologically associated domain at the clustered protocadherin locus. They show that SETDB1 prevents excess CTCF binding and is important for maintaining developmentally important higher-order chromatin organization.
