Polarity signaling ensures epidermal homeostasis by coupling cellular mechanics and genomic integrity

Many developing tissues require Par-driven polarization, but its role in mammalian tissue maintenance is unclear. Here, the authors show that in mouse epidermis, Par3 governs tissue homeostasis not via orientation of cell division but by coupling cell mechanics with mitotic accuracy and genome integrity.

Tracheal tuft cells release ATP and link innate to adaptive immunity in pneumonia

Tracheal tuft cells have been shown to facilitate the recruitment of immune cells during infection of the airways. Here the authors show that P. aeruginosa lung infection in mice activates tuft cells to release ATP which subsequently activates DC and promotes IL-17A secreting T cells.

JAM-A regulates cortical dynein localization through Cdc42 to control planar spindle orientation during mitosis

Polarized epithelial cells orient their mitotic spindles in the plane of the sheet but the role of cell adhesion molecules in this process is poorly understood. Here Tuncay et al. show that JAM-A regulates spindle orientation by creating a gradient of PtdIns(3,4,5)P₃, regulating cortical actin assembly and localizing dynactin to the cell cortex.

mTORC1 and mTORC2 regulate skin morphogenesis and epidermal barrier formation

mTOR regulates cell growth via a protein complex including mTORC1 and mTORC2, but their role in skin morphogenesis is unclear. Here, the authors delete mTORC1 and mTORC2 from the epidermis and see epidermal deficiencies but both mTORCs play distinct roles in skin morphogenesis.
