Single-cell ATAC-seq is limited by cost and scalability. Here, the authors present IT-scATAC-seq, a semi-automated, cost-effective method using indexed Tn5 transposomes, enabling high-fidelity, high-throughput chromatin accessibility profiling in developmental and immune contexts.
