Cyclic dinucleotides are conserved second messengers but current detection methods are limited. Here the authors engineer a Förster resonance energy transfer (FRET) based biosensor, BioSTING, which gives real-time in vitro detection of these nucleotides.

SLC19A1 transports immunoreactive cyclic dinucleotides

A genome-wide CRISPR-interference screen is used to identify the reduced folate carrier SLC19A1 as the major transporter of cyclic dinucleotides in human cells, with potential roles in immunotherapeutic treatment of cancer, immune responses to pathogens and inflammatory diseases.

The helicase DDX41 recognizes the bacterial secondary messengers cyclic di-GMP and cyclic di-AMP to activate a type I interferon immune response

Activation of type I interferon by c-di-GMP and c-di-AMP depends on the adaptor STING. Cheng and colleagues show that these bacterial secondary messengers are detected by the helicase DDX41, which forms a complex with STING.

A TRAF3-NIK module differentially regulates DNA vs RNA pathways in innate immune signaling

The innate immunity system detects viral pathogens by sensing viral DNA or RNA via distinct pathways, but whether these pathways cross-regulate is unclear. Here the authors show that TRAF3, a known regulator of the RNA-sensing pathway, modulates an NF-κB activator NIK to control DNA-sensing by the adaptor STING in immune cells.
