Lysosomes are the key digestive organelles of differentiated keratinocytes in the epidermis. Mahanty et al. show that ER stress but not mTOR-MiT/TFE factors promotes lysosome biogenesis during keratinocyte differentiation, which is critical for epidermal homeostasis.
