Solenoid proteins are elongated tandem repeat proteins with diverse biological functions, however, modular repeat architecture presents challenges for de novo protein design. Here, the authors develop an in silico evolution platform integrating a solenoid discriminator network and AlphaFold2, successfully designing and experimentally validating new α- and β-solenoid structures.
