Molecular dissection of colorectal cancer in pre-clinical models identifies biomarkers predicting sensitivity to EGFR inhibitors

The heterogeneity of colorectal cancer has important clinical and therapeutic implications. Here the authors analysed the responses of a large biobank of organoids and xenografts derived from colorectal patients to a panel of clinically relevant therapeutic agents to identify genes signatures associated with drug response.

Improved binding site assignment by high-resolution mapping of RNA–protein interactions using iCLIP

Individual-nucleotide resolution crosslinking and immunoprecipitation (iCLIP) can map RNA binding sites of RNA-binding proteins (RBPs). Here, the authors report an analysis tool that improves the binding site assignment for some RBPs that have length-dependent broader distribution for their iCLIP fragments.

A brave new world of RNA-binding proteins

Recent proteome-wide studies have uncovered hundreds of RNA-binding proteins (RBPs) that lack conventional RNA-binding domains. These RBPs instead use intrinsically disordered regions, protein–protein interaction interfaces and enzymatic cores to bind RNA. Interestingly, some RBPs are regulated by RNA rather than regulate RNA.
