Disruption of lysosomal proteolysis in astrocytes facilitates midbrain organoid proteostasis failure in an early-onset Parkinson’s disease model

The protein DJ1, encoded by the PARK7 gene, is causally linked to development of early-onset PD. Here the authors observed that the loss of DJ1 function in midbrain organoids led to astrocyte dysfunction, impairing protein clearance, accumulation of α-synuclein.

A pesticide and iPSC dopaminergic neuron screen identifies and classifies Parkinson-relevant pesticides

Parkinson’s disease (PD) is linked to environmental factors. Through quantitative epidemiology, this study ties 53 pesticides to PD. An innovative human stem cell platform revealed that 10 of these were directly toxic to human dopamine neurons.

Programming human pluripotent stem cells into white and brown adipocytes

Cowan and colleagues have developed a method to efficiently differentiate human pluripotent stem cells into functional white or brown adipocytes, through the transient expression of PPARG2 alone or in combination with CEBP and PRDM16. The programmed cells are able to give rise to ectopic fat pads with white or brown adipose tissue characteristics.

From noncoding variant to phenotype via SORT1 at the 1p13 cholesterol locus

A non-coding polymorphism at a locus associated with myocardial infarction in humans creates a CCAAT/enhancer binding protein transcription factor binding site and alters the hepatic expression of the SORT1 gene. These authors show that modulating Sort1 levels in mouse liver alters levels of plasma low-density lipoprotein cholesterol and very low-density lipoprotein, potentially explaining why polymorphisms at this locus are associated with heart disease.

Differentiation stage determines potential of hematopoietic cells for reprogramming into induced pluripotent stem cells

Konrad Hochedlinger and colleagues show that hematopoietic stem and progenitor cells give rise to induced pluripotent stem cells 300 times more efficiently than terminally differentiated B and T lymphocytes, yielding reprogramming efficiencies of up to 27%. These findings identify adult hematopoietic progenitors as an attractive cell type for applications of iPS technology in research and therapy.
