AB₅ subtilase cytotoxin inactivates the endoplasmic reticulum chaperone BiP

Shiga toxigenic Escherichia coli can cause gastrointestinal disease in humans, with potentially life-threatening consequences. This paper elucidates the mechanism of the bacteria-induced host cell death; a protein crucially involved in endoplasmic reticulum function is specifically targeted and inactivated by the toxin's protease subunit.

Targeting of a natural killer cell receptor family by a viral immunoevasin

The mode by which NK cell receptors are bound to their ligands has been unclear. Rossjohn and colleagues show that the cytomegalovirus immunoevasin m157 binds the NK cell receptor Ly49 by its stalk region and not via the expected membrane-distal lectin domain.

T-cell receptors expressed on mucosal-associated invariant T cells function in a similar manner to innate immune receptors by recognizing small molecules such as microbial metabolites. Incorporation of a non-human glycan mediates human susceptibility to a bacterial toxin

Shiga toxigenic Escherichia coli causes severe gastrointestinal disease, which is in part mediated by subtilase cytotoxin. The B subunit of this toxin is now shown to have high affinity to glycans containing N-glycolylneuraminic acid, a saccharide that is not synthesized by humans. Instead it is ingested by dietary intake of red meat and dairy products and subsequently incorporated into intestinal and kidney tissue.

A mortise–tenon joint in the transmembrane domain modulates autotransporter assembly into bacterial outer membranes

Bacterial autotransporters are folded in a process that entraps segments of their N-terminal passenger domain. Here, Leyton et al. identify glycine-aromatic mortise and tenon motifs that mediate the passenger domain’s translocation to the bacterial cell surface, and show that the motifs are evolutionarily conserved.

The oligomeric assembly of galectin-11 is critical for anti-parasitic activity in sheep (Ovis aries)

Sakthivel et al. report two natural genetic variants of galectin-11 in sheep and demonstrate that their ability to dimerise correlates with the differences in their anti-parasitic activity that triggers larval arrest. This study provides a basis for genetic studies toward selective breeding of animals to improve their resistance to parasites.
