How synovial fibroblasts activate cartilage degradation in arthritis is not well understood. Here, the authors identify an O-glycosylation program and glycosylated Calnexin in synovium’s edge fibroblasts as drivers of extracellular matrix degradation, highlighting potential therapeutic targets.
