T-Plastin reinforces membrane protrusions to bridge matrix gaps during cell migration

In vivo, cells migrate across a diverse landscape of extracellular matrix containing gaps which present a challenge for cells to protrude across. Here, the authors show that T-Plastin strengthens protrusive actin networks to promote protrusion, extracellular matrix gap-bridging, and cell migration.

Bacteria assemble the surface layer (S-layer), a crystalline protein coat surrounding the curved surface, using protein self-assembly. Here authors image native and purified RsaA, the S-layer protein from C. crescentus, and show that protein crystallization alone is sufficient to assemble and maintain the S-layer in vivo.

Large single-molecule fluorescence enhancements produced by a bowtie nanoantenna

A 1,340-fold increase in single-molecule fluorescence has been observed from a lithographically fabricated gold bowtie nanoantenna — approximately an order of magnitude greater than that achieved in previous reports on such structures. The improvement results from an estimated ninefold increase in quantum efficiency, caused by enhanced absorption and an increased radiative emission rate.

Super-resolution imaging in live Caulobacter crescentus cells using photoswitchable EYFP

Previous work showed that the commonly used fluorescent protein EYFP can be bleached and reactivated. Exploiting this property allows super-resolution in vivo imaging of EYFP-labeled structures in living bacteria. Fölling et al., also in this issue, describe a related approach for super-resolution imaging using other ordinary fluorophores.

Watching conformational- and photodynamics of single fluorescent proteins in solution

A method for observing the photodynamics of single molecules, without having to immobilize them to a surface or confine them within vesicles, has been used to study the important photosynthetic antenna protein, allophycocyanin. In an interesting demonstration of how bacterial subcellular organization influences physiology, polar accumulation of PopZ protein in a membraneless microdomain is found to drive asymmetric phosphorylation of CtrA-P, which creates a gradient that is responsible for asymmetric cell division in Caulobacter.
