PXR regulates glycolipid metabolism, but the role of intestinal PXR in metabolic diseases remains unknown. Here, the authors show the intestinal-selective PXR activation regulates B3galt5 expression and maintains metabolic homeostasis via regulating the intestinal mucus barrier.
