Microsatellite-stable/proficient mismatch repair colorectal cancers are resistant to immune checkpoint blockade immunotherapy due to their immune suppressive tumour microenvironment. Here the authors show in mouse models that the transcription factor Arid3b contributes to the suppression of CD8 + T cell infiltration and activity in these tumours via suppressing Runx3, a known regulator of tissue resident memory phenotype in CD8 + T cells.
