The Ca²⁺ sensor STIM1 regulates the type I interferon response by retaining the signaling adaptor STING at the endoplasmic reticulum

STIM1 is a calcium sensor that is essential for functional lymphocyte responses. Gwack and colleagues demonstrate a calcium-independent role for STIM1 in macrophages that regulates their production of type I interferons.

Orai inhibition modulates pulmonary ILC2 metabolism and alleviates airway hyperreactivity in murine and humanized models

The regulation and intracellular transport of Ca²⁺ in immune cells involves Ca²⁺ release-activated Ca²⁺ (CRAC) channels. Here the authors show targeting CRAC components Orai1 and Orai2 modulates pulmonary ILC2 cells altering their metabolism, function and is linked to alleviation of immunopathology in a murine model of allergic airway disease.

Insulin-activated store-operated Ca²⁺ entry via Orai1 induces podocyte actin remodeling and causes proteinuria

Perturbations of Ca²⁺ signaling in podocytes may deteriorate kidney function and eventually lead to proteinuria. Here the authors show that insulin can affect the function of the calcium regulator Ora1 in podocytes, which is critical for maintaining kidney filter integrity.

A novel EF-hand protein, CRACR2A, is a cytosolic Ca²⁺ sensor that stabilizes CRAC channels in T cells

The calcium-binding protein CRACR2A positively regulates the activity of CRAC channels. CRACR2A is a ternary complex which binds to, and stabilizes, the interaction between STIM1 and Orai1, and disassociates as intracellular calcium levels rise.
