AAK1 activation-mediated iron trafficking drives ferroptotic cell death

Endocytosis of TFR1 plays a crucial role in extracelluar iron uptake. Here the authors revealed that PKCβII facilitates the endocytosis of TFR1 and increases cellular iron levels through the phosphorylation of AAK1, thus promoting ferroptosis of tumor cells.

Galectin-13 reduces membrane localization of SLC7A11 for ferroptosis propagation

Cells undergoing ferroptosis secrete Galectin-13, which binds to CD44 and inhibits the plasma membrane localization of SLC7A11 in neighboring cells, thereby accelerating neighboring cell death and promoting ferroptosis propagation.

PKCβII phosphorylates ACSL4 to amplify lipid peroxidation to induce ferroptosis

Through CRISPR–Cas9 and kinase inhibitor screening, Zhang et al. show that PKCβII phosphorylates and activates ACSL4 to enhance polyunsaturated fatty acid-containing lipid biosynthesis, thereby promoting accumulation of lipid peroxidation and ferroptosis.
